Cell culture conditions for ChIP-exo
Cell cultures needs to be grown overnight before starting the ChIP-exo protocol. Cells should be incubated directly from -80 °C stock using a minimal medium plus the desired carbon source and selective drug, such as kanamicine.
After growing overnight, the culture needs to be diluted in the same medium using a final volume of 60 mL and to an optical density at 600 nm (OD600) of 0.02. In order to achieve the maximal performance of the technique, cells need to be in the mid-log growth phase. Normally, that phase is reached when OD600=0.4.
