pMA7 plasmid
Plasmid pMA7 includes a bet gene and a Dam, controlled by an arabinose-inducible (PBAD) promoter. The bet gene encodes the Beta protein which required for ssDNA hybridization and stabilization. The dam gene encodes the Dam (DNA adenine methylase). The Dam is hypothesized to generate significant methylation of the newly synthesized, disabling detection of incorporated mismatches by use of mismatched repair (MMR) system and thus reducing the rate of off-target mutation.
