Basic steps of ELISA
A wide variety of ELISA types are available for different applications. However, all ELISA types still require the same basic elements:
- Coating/capture: Immobilization of the antibody or antigen to the surface of the microplate wells
- Blocking: Addition of irrelevant protein or other molecules to cover all unsaturated surface-binding sites of the microplate wells
- Probing/detection: Incubation with the labeled antigen or antibody
- Signal measurement: Detection of the signal generated via the labeled antigen or antibody
